α-Acetolactate Decarboxylase is an enzyme that effectively reduces diacetyl content in beer and improves its flavor. It directly converts the diacetyl precursor, acetolactate, produced during beer fermentation into acetoin, bypassing the formation of the off-flavor compound diacetyl. This speeds up beer maturation, shortens production cycles, and increases equipment utilization, offering significant economic and social benefits.

Production Process of α-Acetolactate Decarboxylase

Produced through fermentation:

The production method uses integrated recombinant Bacillus subtilis as the strain. The process involves fermenting in a nutrient medium to produce α-acetolactate decarboxylase, detailed as follows:

  1. Recombinant Strain: The integrated recombinant Bacillus subtilis has the α-acetolactate decarboxylase expression element integrated into its chromosome, selected to obtain the recombinant strain.
  2. Nutrient Medium: A liquid medium containing 0.1-5% trypticase peptone, 0.1-5% yeast extract, and 0.5-2% NaCl, or a similar protein content hydrolyzed casein medium instead of trypticase peptone.
  3. Production Method: The production includes inoculation, cultivation, separation, concentration, and packaging. The process involves:
    1. Using the constructed recombinant Bacillus subtilis strain.
    2. Fermenting at 30-40°C, pH 6.0-8.0, under aerobic conditions with an inoculum of 0.5%-15%.
    3. Aerobic fermentation for 18-48 hours.
    4. Centrifugation at 400-16,000g or filtration through a 0.2-0.6 μm membrane to remove bacteria.
    5. Concentrating the supernatant using a 5000-10,000 molecular weight cutoff ultrafiltration membrane to obtain the final product.
    6. Store at 4-10°C.

α-Acetolactate Decarboxylase (ALDC, EC4.1.1.5) is widely used by breweries as a maturation aid.

Storage Conditions: Keep sealed, protected from light, and store in a dry, cool, well-ventilated place.

Packaging: Bulk packaging in 25 kg cardboard drums; samples in 1 kg aluminum foil bags, or customized packaging as per customer requirements.
